Materials and
Methods:
Medaka
embryos were supplied by the Biology Department of Franklin
& Marshall College. They were harvested and separated by
Dr. Richard Fluck. The cyclopamine was the kind gift of Dr.
William Gaffield (USDA).
Treated
Embryos:
* Embryos were treated at
30% and 70% epiboly.
* Treated with 200
mM1solution
of cyclopamine (dissolved in 95% ethanol). 0.4 mL of
cyclopamine, that had previously been dissolved in 95%
ethanol was added to 4.6 mL of instant ocean.
*Treated
with 100 mM
solution of cyclopamine, after embryos of previous treatment
all died. 0.2 mL of cyclopamine dissolved in 95% ethanol was
added to 4.8 mL of instant ocean.
Control
Embryos:
* Treated with solution consisting of 0.4 mL of 95%
ethanol and 4.6 mL of instant ocean when the 200
mM
cyclopamine treatment was used.
*
Treated with solution consisting of 0.2 mL of 95% ethanol
and 4.8 mL of instant ocean when the 100 mM
cyclopamine treatment was used.
